Regina Casadesus came from a highly musical family. Her father was the conductor Luis Casadesus (1850-1919) and several of her siblings were musicians. She married the author and anarchist Aurèle Patorni (1880-1955) in 1906.

Her career was stronly connected with the Société des Instruments Anciens that was founded by her brother Henri. She participated in 75 tours during a 35 years. Paris, Berlin, Moscow, Brussels, Madrid, London and Vienna were among the cities where she performed.

She died in 1961 and in 1962 her autobiography "Ma famille Casadesus: souvenirs d'une claveciniste 1890-1955" was published.
