Delphine Ugalde was the daughter of the music publisher Claude Beaucé. Her mother was a musician and she taught her the piano. When she was nine years old, she gave a recital at the Salle Herz. When she was sixteen years old, she married the musician and piano maker Casto de Ugalde, who was known as Fréry. They had a child that died prematurely.

She continued her studies with Théodore-François Moreau-Sainti. Shortly after Adolphe Adam contracted her for the Théâtre-National, but then the Revolution of 1848 broke out. She was offered an engagement in Madrid, where he sang Verdi's music accompanied by her husband. Later in 1848 she debuted at the Opéra Comique. On 3 January 1849 she was the first Virginie in Ambroise Thomas' "Le Caïd" and on 18 May 1849 the first Nerilha in Fromental Halévy's "La Fée aux Roses". In 1851 she sang in London in Auber's " L'Enfant prodigue".

After a voice illness she was about to make her comeaback at the Théâtre Favart, when Caroline Miolan-Carvalho suddenly fell ill. She replaced her that evening as Isabelle in Hérold's "Le Pré aux Clercs" after studying the role for only a few hours. On 26 January she was in the premiere of Ambroise Thomas' "Psyché". Her husband died in 1858 and at that time she left the Opéra-Comique to give concerts in London. Back in Paris she was contracted by the Théâtre-Lyrique. In 1860 she returned to the Opéra-Comique once more.

In 1862 she joined the Théâtre des Bouffes-Parisiens and in September 1866 she became its director until July 1867. Further engagements followed until she retired from the stage in 1871. She concentrated on teaching and Marie Sasse was one of her students. From 1885 to 1888 she directed the Théâtre des Bouffes-Parisiens again. Her second husband François Varcollier was its co-director.

She died in 1910 in Paris, and she was buried at the Montmartre Cemetery. Her daughter Marguerite Ugalde and her granddaughter Jeanne Ugalde were actors as well.
