Peppiono de Filippo was the son of playwright Eduardo Scarpetta. He started acting in his childhood. In 1931 he formed the company "Compagnia Teatro Umoristico: i De Filippo" with his brother Eduardo and his sister Titina. They toured all over Italy with their comedies.
In 1944 he fell out with Eduardo and left the company and successfully established a career in the movies. He also became a well known television personality and for television he created the personality Pappagone.

He was married three times. His first wife was the actress Adelina Carloni, with whom he was married from 1929 to 1945. Their son Luigi de Filippo (b.1930) also became an actor.
