Lucien Muratore was born in Marseille as the son of immigrants from Piedmont. He started his acting career at an early age at the Théâtre des Variétés in Marseille. In Paris he appeared with Réjane and Sarah Bernhardt at the Théâtre de l'Odéon. In Marseille he studied music at the Conservatory and in Paris Edmond Vergnet was his teacher. He started as a bariton but over time he became a tenor.

On 16 December 1902 he made his debut at the Opéra-Comique in Paris in 'Reynaldo Hahn's "La carmelite". His first appearance at the Opéra in Paris was in 1905. He had married the opera singer Marguerite Bériza and they made several Edison cylinder recordings together in 1906/1907.

Muratore sang many important parts in Paris, including Don José in "Carmen". In 1909 he performed in Bordeaux and in 1911 in Monte Carlo. He had left his wife and live with the prima ballerina Marceline Rouvier. They had a daughter, Ariana. Mutatore left her soon afterwards for Lina Cavalieri and in 1913 he divorced Marguerite and married Lina in the same year. From then on, he mostly lived in the USA. Between 1914 and 1916 he and Lina they appeared together in several silent movies. In 1919 he separated from Cavalieri and in 1927 they divorced.

In 1931 he ended his stage career in Fauré's "Penelope". after that he taught singing, and he worked as a director. Kenneth Neate (1914-1997) was his pupil and Muratore gave him several costumes that he had worn for the part of Don José. From 1929 to 1934 he was mayor of Biot, Alpes-Maritimes. In April 1944 he was appointed director of the Opéra-Comique in Paris. This engagement ended on 1 September 1944 after the liberation of Paris.

Muratore died in 1954 is Paris and he was buried in Marseille. Antoine Sartorio created the sculpture on his tomb.
