Fernand Desmoulin studied medicine when he realised that he wanted to be a painter. He abandoned his medical studies and William Bouguereau, Luc-Olivier Merson and Félix Bracquemond taught him painting and engraving. He exhibited at the Salon des artistes français from 1883 to 1890 and at the Société nationale des beaux-arts from 1891 to 1899 and from 1906 to 1910.

Through his contributions for "La Vie Moderne" he befriended Georges Charpentier and from 1887 Émile Zola. His wife Gabrielle Génie died in 1894. He found a new partner, but they separated in 1899. He felt depressed and he embraced spiritualism. Between 1900 and 1902 he produced mediumistic drawings that were exhibited in 1901. In 1905 he married Anna van Oosterom, the granddaughter of Heinrich Bürger (1806-1858).

In 1897 Desmoulin received the Legion of Honour and in 1903 he was promoted to Officer of that order. He died in 1914 in Venice and he was buried at the Montmartre cemetery in Paris.
