Ernestine Boie was the daughter of a preacher from Meldorf who later became provost of Flensburg. She was the younger sister of the poet and publisher Heinrich Christian Boie (1744-1806). Her brother knew Johann Heinrich Voss well and after a long correspondence they met in 1774. In 1777 his financial position allowed them to marry. Their marriage was harmonious they had five children. Like her husband she worked as a writer.

They lived in Jena from 1802 to 1805 and she got along very well with Charlotte von Schiller and Johann Wolfgang von Goethe who visited them frequently at the time. Her husband died in 1826 and together with their son Abraham she annotated and published many of his letters. In 1831 she privately published a memoire of Emilie Heins (c1776-1831). After her death in 1834 several of her essays and poems were published.
