Otto Reuchlin was born into a family of entrepeneurs. From an early age he worked as a merchant, trading fertilizers and wine. In 1871 he founded the company Plate, Reuchlin & Co together with Antoine Plate. This company became the Nederlandsch-Amerikaansche Stoomvaart-Maatschappij (NASM) in 1873 and later is was known as the Holland-America Line (HAL). The company transported emigrants from Europe from the harbour of Rotterdam to the USA. Plate left the company in 1880, but Reuchlin only retired from the management in 1919.

On 27 April 1871 he married Carolina Helena Schumacher (b.1849). Their second son Johan George was one of the three Dutch people that died after the Titanic sank in 1912. In 1890 he received the French Légion d'honneur. In the Netherlands he was decorated as well. After Carolina's death around 1897 Reuchlin married her sister.
