James Buchanan was the son of the farmer George Buchanan from Drymen, Dumbartonshire. He originally worked as a linen trader, and he enrolled at the Merchants House in Glasgow in 1790. He and his cousin Moses Steven were partners in the West India merchant firm Buchanan, Steven & Co and later in its successor Dennistoun, Buchanan & Co. This firm was a subscriber to the pro-slavery Glasgow West India Association.

Buchanan was an early investor in The Glasgow Banking Company, a predecessor of Lloyds. He married Ellison McCallum (1770-1842) in 1801. At the time of his death, he was probably one of the wealthiest men in Scotland. His portrait was painted by John Graham Gilbert (1794-1866).
