Enrico Pollastrini was first educated by Vincenzo De Bonis in Livorno. In Florence he was a pupil of Giuseppe Bezzuoli and Pietro Benvenuti. He was appointed director of the Academy of Fine Arts in Florence in 1867, succeeding Tomasso Gazzarrini. He held this office until 1875.

He died in 1876 in Florence and he was buried in the Famedio of the Sanctuary of Montenero near Livorno. His work can now be found at the museo civico Giovanni Fattori in Livorno. His best known Picture "Gli esuli di Siena" was lost during the Second World War.
